干扰素基因刺激蛋白 (STING, TMEM173, MITA) 是一种跨膜接头蛋白,它是细胞病原细胞浆 DNA 的天然免疫应答的一个关键组分 【1,2】。STING 是一种被发现主要在内质网中普遍表达的蛋白 【1】。在细胞浆 DNA 的刺激下,酶 cGAMP 合成酶 (cGAS) 会产生第二信使环-GMP-AMP (cGAMP) 【3,4】。cGAMP 结合并激活 STING 【3,4】。此外,通过 DDX41 和 IFI16 等核酸感应蛋白检测细胞浆 DNA 会导致 STING 激活 【5,6】。激活后,STING 与 TBK1 转位至核周内体,并在 Ser366(小鼠内为 Ser365)位点被 ULK1 磷酸化【7, 8】。TBK1 激酶磷酸化并激活 IRF-3 和 NF-κB,从而诱导 I 型干扰素和其他免疫应答基因 【1,2,7】。
